A PEST sequence is a peptide sequence that is rich in proline (P), glutamic acid (E), serine (S), and threonine (T). It has been confirmed that PEST fused at firefly luciferase at C-terminal significantly reduced the intracellular half-life of luciferase, making it more suitable to faithfully monitor rapid yet subtle changes in transcription in a medium to high throughput format.
