The mitochondrion plays a vital role in both apoptotic and necrotic cell death. The mitochondrial permeability transition pore (mPTP) is a nonspecific channel formed by components from the inner and outer mitochondrial membranes and is involved in the release of mitochondrial components during cell death.
The Mitochondrial Permeability Transition Pore (mPTP) Imaging Assay Kit is based on established experimental methods for detecting mitochondrial transition pore opening. This assay employs calcein AM—a colorless, nonfluorescent esterase substrate—and CoCl₂, a quencher of calcein fluorescence, to selectively label mitochondria.
Key Features
• Direct mPTP Measurement: Provides a direct method for assessing mitochondrial permeability transition pore opening.
• Selective Mitochondrial Labeling: Uses calcein AM (fluorescent upon esterase cleavage) and CoCl₂ (quenches cytosolic fluorescence) to specifically label mitochondria.
• Cytosolic Signal Suppression: CoCl₂ effectively quenches cytosolic calcein fluorescence, eliminating background signal.
• Easy Workflow: Simple cell loading, quenching, and detection steps without complex instrumentation.
